General description

Anti-von Willebrand Factor is produced in rabbit using purified von Willebrand factor (factor VIII R:Ag) as the immunogen.
Human von Willebrand factor (factor VIII R:Ag) is a 270 kDa multimeric plasma gylcoprotein. The von Willebrand factor has functional binding domains to platelet glycoprotein Ib, glycoprotein IIb/IIIa, collagen and heparin. The factor is synthesized by endothelial cells and is also present in platelets and megakaryocytes.

Specificity

The antibody specifically reacts with the cytoplasm of human endothelial cells from normal and neoplastic blood and from lymphatic vessels. It also reacts with human endocardium, platelets and megakaryocytes. It may be used for immunohistological studies of the vascular structures in frozen tissue preparations and enzymatically pre-digested, formalin, methacarn, Bouin′s or ethanol-fixed and paraffin-embedded tissue sections.

Application

Anti-von Willebrand Factor antibody produced in rabbit has been used for:
  • western blot analysis and immunostaining
  • immunocytochemistry
  • immunohistochemistry
  • indirect immunofluorescence

Biochem/physiol Actions

It mediates platelet adhesion to injured vessel walls and serves as a carrier and stabilizer for coagulation factor VIII.

Physical form

Solution in 0.01 M phosphate buffered saline, pH 7.4, containing 15 mM sodium azide

Preparation Note

Whole antiserum is purified to provide an IgG fraction of antiserum.
