Rat pituitary adenoma

Cell Line Description

RC-4B/C1 is a clone of the parent RC-4B/C which was established from a 3 year-old CAI/FMaixF344/FMai(F1) male rat pituitary adenoma. Staining studies have shown the cell line contains many bi-hormonal cells with FHSß, LHß and prolactin. Electron microscopy shows the appearance of endogenous C-type rat retroviral particles.

Application

Study of gonadotrophic functions of pituitary gland and regulation of pituitary gonadotrophin gene expression.

Culture Medium

DMEM:EMEM (EBSS) (1:1) + 2mM Glutamine + 0.2mg/ml BSA + 0.5% Non Essential Amino Acids (NEAA) + 5% Nu-Serum (Collab)+ 2ng/ml EGF.

Subculture Routine

Split sub-confluent cultures (70-80%) 1:6 i.e. seeding at 2x10,000 cells/cm2 using 0.25% trypsin/EDTA; 10% CO2; 34°C. Saturation density is reached at 1x 100,000 cells/cm2. Cells may attach lightly and so handling should be kept to a minimum. Care shoul
