High affinity nerve growth factor receptor (UniProt: P04629; also known as EC:2.7.10.1, Neurotrophic tyrosine kinase receptor type 1, TRK1-transforming tyrosine kinase protein, Tropomyosin-related kinase A, Tyrosine kinase receptor, Tyrosine kinase receptor A, Trk-A, gp140trk, p140-TrkA) is encoded by the NTRK1 (also known as MTC, TRK, TRKA) gene (Gene ID: 4914) in human. Trk-A is a single-pass type I membrane protein that is synthesized with a signal peptide (aa 1-32), which is subsequently cleaved off to produce the mature protein that contains an extracellular domain (aa 33-423), a transmembrane domain (aa 424-439), and a cytoplasmic domain (aa 440-796). The extracellular domain of Trk-A is composed by five domains: a leucine-rich region, two cysteine-rich domains and two immunoglobulin (Ig)-like domains. Trk-A serves as a receptor tyrosine kinase involved in the development and the maturation of the central and peripheral nervous system through regulation of proliferation, differentiation, and survival of sympathetic and nervous neurons. It serves as a high affinity receptor for NGF, which is its primary ligand. It can also bind and be activated by neurotrophin-3 (NTF3). However, NTF3 only supports axonal extension through NTRK1 but does not affect neuron survival. Upon dimeric NGF ligand-binding, it undergoes homodimerization, autophosphorylation. and activation. Clone MNAC13 recognizes Trk-A with high affinity and specificity and displays a neutralizing activity toward the NGF/TrkA interaction. (Ref.: Covaceuszach, S., et al. (2005). Specificity

Clone MNAC13 is a mouse monoclonal antibody that detects TrkA. It targets an epitope within 17 amino acids within the first Ig-like C2 type domain from the extracellular domain.

Immunogen

BALB/C 3T3-transfected cells expressing human Trk-A protein.

Application

Quality Control Testing

Evaluated by ELISA with recombinant Human TrkA.

ELISA Analysis (ELISA): Various dilutions of this antibody detected recombinant Human TrkA.

Storage and Stability

Store at -10°C to -25°C. Handling Recommendations: Upon receipt and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance.
