Specificity

Clone CT3 is a mouse monoclonal antibody that detects Glypican-2 (GPC2) and its glycated forms. It targets an epitope within the C-terminal region.

Immunogen

KLH-conjugated linear peptide corresponding to 50 amino acids from the C-terminal region of human Glypican-2 (GPC2).

Target description

Glypican-2 (UniProt: Q8N158; also known as GPC2) is encoded by the GPC2 gene (Gene ID: 221914) in human. Glypicans belong to a family of heparan sulfate proteoglycans that are attached to the cell membrane via a GPI-anchor. They interact with multiple ligands, including morphogens, growth factors, chemokines, ligands, receptors, and components of the extracellular matrix through their heparan sulfate chains and core protein. GPC2 is synthesized with a signal sequence (aa 1-23), which is subsequently cleaved off to produce the mature GPC2 (aa 24-554). It also contains a propeptide sequence (aa 555-579) that is removed in the mature form. Glypicans are reported to regulate Wnt/β-catenin signaling and affect cell growth and proliferation. Overexpression of GPC2 is reported in several pediatric cancers. Its higher expression is also shown in neuroblastoma compared to the normal peripheral nerve tissue. GPC2 can modulate Wnt signaling and regulate MYCN in neuroblastoma pathogenesis. Genetically silencing of CPC2 is shown to inhibit neuroblastoma cell growth by suppressing N-Myc expression. Clone CT3 specifically binds to GPC2 in human cancer cells and does not react with normal tissues. It recognizes tumor-associated exons 3 and 10 of GPC2. (Ref.: Li, N., et al. (2020). J. Histochem. Cytochem. 68(12); 841-862; Li, N., et al., 2021, Cell Rep. Medicine 2, 100297).

Physical form

Purified mouse monoclonal antibody IgG1 in PBS without azide.

Reconstitution

0.5 mg/mL. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Store at -10°C to -25°C. Handling Recommendations: Upon receipt and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance.

Other Notes

Concentration: Please refer to the Certificate of Analysis for the lot-specific concentration.
