β Galactosidase, also known as Beta-galactosidase, Beta-gal, LacZ, or Lactase, and encoded by the gene lacZ, is the enzyme in bacteria that hydrolyzes galactosides (sugars chains containing galactose) in to monosaccharides. β Galactosidase is typically an extracellularly produced enzyme that hydrolyzes the beta-glycosidic bond between galactose and another organic moiety. In E. coli, the β Galactosidase gene is present as part of the inducible lac operon system which is activated in the presence of lactose when glucose level is low. Today, bacterial beta galactosidase is used in molecular biology as a reporter marker to monitor gene expression, and because the enzyme has two functional domains, it forms the basis for α-complementation which is the methodology behind for the blue/white screening of recombinant clones. The presence or absence of an active β-galactosidase may be detected by X-gal, which produces a characteristic blue dye when cleaved by β-galactosidase, thereby providing an easy means of distinguishing the presence or absence of cloned product in a plasmid.
