General description

Gamma-secretase is a protein complex containing presenilin, nicastrin, APH-1, and PEN-2. The leupeptin analogue Z-Leu-Leu-Nle-al (N-benzyloxycarbonyl-Leu-Leu-norleucinal; Benzyloxicarbonyl-Leu-Leu-Nle-CHO (LLNle)) and closely related peptides are used as protease and a gamma secretase inhibitors.

Application

γ Secretase Inhibitor has been used to study the suppressive miR-129 functions through inhibition on estrogen receptor 1 (ESR1) and NOTCH signaling in breast cancer stem cells.[1]

    1. The solubility of this product is in DMSO at 1 mg/ml or higher.

      In biological experiments, water is the most preferred solvent. However, several organic compounds are either not soluble in water or degrade rapidly in the presence of moisture. If DMSO is a recommended solvent, it is best to use a fresh stock bottle that is deemed free of any moisture. Any contaminating moisture may accelerate the degradation of the compound in question or may render it insoluble. It is best to make the initial serial dilutions only in DMSO and then add the final diluted sample to your buffer or the incubation medium. The final concentration of DMSO in vivo should not exceed more than 0.1 %.
