Specificity

Clone 1K16 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects FUN14 domain-containing protein 1 (FUNDC1). It targets an epitope within 20 amino acids from the C-terminal, second cytoplasmic domain.

Immunogen

KLH-conjugated linear peptide corresponding to 20 amino acids from the second cytoplasmic domain of human FUN14 domain-containing protein 1 (FINDC1).

Application

Quality Control Testing

Evaluated by Western Blotting in L6 cell lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected FUNDC1 in L6 cell lysate.

Tested applications

Affinity Binding Assay: A representative lot of this antibody bound FUNDC1 with a KD of 1.5 x 10-7 in an affinity binding assay.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected FUNDC1 in human cerebral cortex tissue sections.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected FUNDC1 in L6 cells.

Anti-FUNDC1, clone 1K16 ZooMAb®, Cat. No. ZRB1282, is a recombinant Rabbit monoclonal antibody that specifically targets FUNDC1 and is tested for use in Affinity Binding Assay, Immunocytochemistry, Immunohistochemistry (Paraffin), and Western Blotting.

Target description

FUN14 domain-containing protein 1 (UniProt: Q8IVP5; also known as FUNDC1) is encoded by the FUNDC1 gene (Gene ID: 139341) in human. FUNDC1 is a multi-pass membrane protein with two cytoplasmic domains, three transmembrane domains, and two mitochondrial intermembrane domains. It is widely expressed and is conserved from Drosophila to human. It is localized exclusively in the mitochondria and acts as an activator of hypoxia-induced mitophagy, which contributes to the mitochondrial quality control. It interacts with LC3 family proteins via its LC3 binding motif YXXL (aa 18-21). Knockdown of endogenous FUNDC1 is shown to inhibit hypoxia-induced mitophagy that can be reversed by the expression of wild-type FUNDC1, but not LC3-interaction-deficient FUNDC1 mutants. Mutations leading to defective YXXL motif abolish its interaction with LC3 and subsequent mitophagy. Phosphorylation of tyrosine 18 in the YXXL motif by Src kinase inhibits the activation of mitophagy. Following hypoxia, tyrosine 18 is dephosphorylated that results in increase in interaction with LC3 proteins and triggers mitophagy. FUNDC1 is shown to reduce the levels of TOM20 and VDAC1 in the outer mitochondrial membrane and TIM23 in the inner membrane without affecting the content of other organelle markers. This reduction can be blocked by treating cells with bafilomycin A1. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb® formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
