General description

Ran GTPase−activating protein 1 (RanGAP1), a guanosine-5′-triphosphatase (GTPase)-activating protein for Ran exists in two different types, where one is cytoplasmic and other is concerted at the cytoplasmic fibers of nuclear pore complexes (NPCs). Mammalian RanGAP1 has a C-terminal extension. Ran GTPase gene is located on human chromosome 22q13.2.

Specificity

Anti-RanGAP1 (N-terminal) recognizes human RanGAP1.

Immunogen

synthetic peptide corresponding to amino acids 1-20 located at the N-terminus of human RanGAP1, conjugated to KLH. This sequence is identical in mouse RanGAP1.

Application

Anti-RanGAP1 (N-terminal) antibody produced in rabbit may be used immunoblotting and immunofluorescence.

Biochem/physiol Actions

Ran GTPase−activating protein 1 (RanGAP1) regulates Ran activity. RanGAP1 can stimulate the GTPase activity of Ran. Ran is also regulated by a chromatin-bound nucleotide exchange factor, regulator of chromosome condensation 1 (RCC1) that keeps Ran in the active guanosine-5′-triphosphate (GTP)-bound state. RanGAP1 has the ability to conjugate to the small ubiquitin-related modifier protein small ubiquitin-related modifier 1 (SUMO-1). RanGAP1 may participate in nuclear protein import.

Physical form

Solution in 0.01 M phos­phate buffered saline, pH 7.4, containing 15 mM sodium azide.

Storage and Stability

For continuous use, store at 2-8 °C for up to one month. For extended storage, freeze in working aliquots. Repeated freezing and thawing, or storage in “frost-free” freezers, is not recommended. If slight turbidity occurs upon prolonged storage, clarify the solution by centrifugation before use. Working dilutions should be discarded if not used within 12 hours.
