General description

Monoclonal Anti- hPlk1 (Polo Like Kinase 1) (mouse IgG2b isotype) is derived from the 35-206 hybridoma produced by the fusion of mouse myeloma cells (PAI) and splenocytes from BALB/c mice immunized with human full length Plk1. Polo-like kinases (Plks) are a family of serine/threonine kinases. The Plk family is structurally and functionally related to the polo gene in Drosophila. Plk1 localizes during mitosis in centrosomes, kinetochores and in the central spindle.

Biochem/physiol Actions

PLK1 is a serine-threonine kinase that is expressed in mitotic cells and cancers. This kinase regulates mitotic entry by modulating cdc2/cyclin B and is also involved in Nlp-mediated nucleation of microtubules .
Polo Like Kinase 1 (Plk1) has a role in tumor progression. Plk1 is a target of the DNA damage checkpoint on the basis that its activity is inhibited by DNA damage in G2 and mitosis. Moreover, Plk1 was found to bind to the sequence-specific DNA binding domain of p53. This interaction between p53 and Plk1 inhibits the transcriptional activation and the proapoptotic functions of p53.

Physical form

Solution in 0.01 M phosphate buffered saline, pH 7.4, containing 15 mM sodium azide.

In animal cells, most microtubules are nucleated at centrosomes. At the onset of mitosis, centrosomes undergo a structural reorganization, termed maturation, which leads to increased microtubule nucleation activity. Centrosome maturation is regulated by several kinases, including Polo-like kinase 1 (Plk1).
