Aplicación

El suero bovino fetal se ha utilizado como suplemento en medios de cultivo celular como el medio Roswell Park Memorial Institute (RPMI-1640) y el medio Eagle modificado de Dulbecco (DMEM)[1] para propiciar el crecimiento de una variedad de líneas celulares.

Acciones bioquímicas o fisiológicas

El suero bovino fetal (FBS) sirve como un suplemento de medios de cultivo celular comúnmente utilizados para cultivos de células, tejidos y órganos in vitro y ex vivo. El FBS sigue siendo un complemento popular para los medios porque aporta una amplia gama de funciones al cultivo celular. El FBS aporta nutrientes, factores de crecimiento y de unión, y protege a las células del daño oxidativo y la apoptosis mediante mecanismos que son difíciles de reproducir en los sistemas de medios sin suero (SFM). Proporciona muchos factores no definidos de promoción del crecimiento y de mejora de la supervivencia de las células en cultivo. Procedente de sangre coagulada, es el suplemento no definido más ampliamente utilizado en cultivos de células eucariotas, en especial de mamíferos. El FBS suele utilizarse con los medios clásicos que se desarrollaron en las décadas de 1950 y 1960, entre ellos muchos medios populares como el medio mínimo esencial (MEM); los medios de Eagle modificados por Dulbecco (DME, DMEM); el medio de Eagle modificado por Iscove (IMDM); el medio M199 y del Instituto Roswell Park Memorial, RPMI-1640.

    1. This product has not undergone testing to determine the presence of dopamine or serotonin.

  3. Could the presence of insoluble precipitates when thawing the serum indicate contamination?

    1. ​Thawed serum may contain precipitates and flocculants due to various reasons, such as residual fibrinogen converting to fibrin upon thawing. Centrifugation at 400 xg can remove excess fibrin, while filtering serum is not recommended to avoid protein depletion and clogged filters.
      Additional points to consider:
      •Cloudiness in human serum products is common and indicates the presence of lipids. Clear, lipid-free human serum products are not available.
      •Microscopic serum precipitates may resemble yeast contamination but are actually lipid aggregates that can be dissolved by lipase. They do not show growth in culture or gram staining and can be removed through centrifugation or filtration.
      •Gel layer or clumps in serum are caused by poor or overheated inactivation or improper mixing of FBS during the Heat inactivation process. Serum with these characteristics should not be used.
      •To minimize denaturation of serum lipoproteins and turbidity, it is recommended to prepare single-use aliquots and avoid repeated freeze-thaw cycles.

  4. Is this product F7524 heat-inactivated? Should it be prior to use?

    1. This product has not been heat inactivated. For most cell culture applications heat inactivation is not necessary. If the application requires inactivation of any heat labile serum components, such as compliment or cytokines, the product may be heat inactivated in the lab. Alternatively, a heat inactivated version of this product is also available. See product F9665.

      To heat inactivate this product:
      1. Allow serum containers to come to room temperature. Swirl bottles of serum immediately before adding to the water bath.
      2. A water-filled control bottles (T1) should be placed in a 56 °C water bath. Place the containers in the water bath up to the serum line. Do not completely submerge the containers. When the temperature of T1 reaches 56 °C, start the timer set for 30 minutes. Do not allow temperatures to exceed 56 °C. Higher temperatures will denature the proteins, gel the serum or alter the performance of the serum.
      3. Gently swirl the bottles every 5 to 10 minutes while in the water bath and check the temperature in the control bottle. It is not uncommon for plastic containers to bow slightly after heat treatment. This is not harmful to the serum.
      4. At the end of 30 minutes remove the serum bottles from the water bath. Aliquot the serum into smaller volumes and freeze. Avoid repeated thaw-freeze cycles.

    1. Calcium levels are not determined for this product. The serum is a natural product that will contain calcium at physiological levels.

    1. Unfortunately, a fetal calf serum that has been specifically tested for cryopreservation of mouse embryos or ovarian mice is not currently offered.   7. How can we heat inactivate the fetal bovine serum cat: F7524? For how long?

    1. All heat inactivated serum products are treated at 56 deg. C for 30 minutes. This process may be applied to manually heat inactivate serum on a per bottle basis using a water bath. When immersing bottles do not allow water to come in contact with the cap/closure.
