Standard procedure and microwave procedure included in package insert.
When treated with periodic acid, glycols are oxidized to aldehydes. After reaction with Schiff′s Reagent (a mixture of pararosaniline and sodium metabisulfite), a pararosaniline adduct is released that stains the glycol-containing cellular elements pink to red. Cellular elements which may be demonstrated with the PAS procedure include glycogen, fungal walls, basement membrane, certain epithelial sulfomucins and sialomucins, neutral mucosubstances, colloid material of the thyroid and the pars intermedia of the pituitary.

  1. How many slides can be stained using the PAS kit (395B-1kt)?

    1 answer
    1. The 395B kit is a kit formulated for multiple uses. Typically, the reagent is only replaced after the staining performance drops off. For kit 395B, calculating an exact range of slides is not possible. The kit is provided with 4 each of the 50 ml bottles of Schiffs reagent and 2 each for the 100 ml bottles for Periodic Acid. Each 50 ml aliquot is probably sufficient for 5-10 uses. The exact number will depend on several factors. If the slides are all to be stained at one time, a larger number of slides could be stained. If the slides are stained 2 or 3 at a time over a large number of weeks, fewer slides could be stained. Again, the solutions are to be replaced as needed and not necessarily after a set number of slides have been stained.

  2. What product do you recommend for Periodic acid, Sodium acetate, and Ammonium hydroxide for PAS staining?

    1. For PAS staining, you can either obtain a kit (395B) or the individual reagents. The kit includes 1% Periodic Acid, Schiff's Reagent, and Gill's 3 Hematoxylin. Below are the various catalog numbers:

      •395B for the PAS kit, with Instructions for Use on the sigmaaldrich.com website.
      •3951-100ml or 395132-1L for the 1% Periodic Acid.
      •3952016-500ml for the Schiff's Reagent. Note that there are other Schiff's reagents for sale, but not all are intended for staining tissue.
      •GHS316-500ml for the Gill's 3 Hematoxylin, available separately from the kit.
