Descripción general

Nile Red is an uncharged hydrophobic molecule and a benzophenoxazone dye.[1][2] It functions as a fluorescent probe for intracellular lipids and hydrophobic domains of proteins. This dye is fluorescent in all organic solvents. Its fluorescence colors range from golden yellow to deep red.[2]

Aplicación

Nile Red has been used for the staining of intracellular lipid droplets[3][4] and neutral lipids.[5][6]

  1. How is shipping temperature determined? And how is it related to the product storage temperature?

    1. Products may be shipped at a different temperature than the recommended long-term storage temperature. If the product quality is sensitive to short-term exposure to conditions other than the recommended long-term storage, it will be shipped on wet or dry-ice. If the product quality is NOT affected by short-term exposure to conditions other than the recommended long-term storage, it will be shipped at ambient temperature. As shipping routes are configured for minimum transit times, shipping at ambient temperature helps control shipping costs for our customers.   2. How can I determine the shelf life / expiration / retest date of this product?

    1. If this product has an expiration or retest date, it will be shown on the Certificate of Analysis (COA, CofA). If there is no retest or expiration date listed on the product's COA, we do not have suitable stability data to determine a shelf life. For these products, the only date on the COA will be the release date; a retest, expiration, or use-by-date will not be displayed.
      For all products, we recommend handling per defined conditions as printed in our product literature and website product descriptions. We recommend that products should be routinely inspected by customers to ensure they perform as expected.
      For products without retest or expiration dates, our standard warranty of 1 year from the date of shipment is applicable.

  3. What kind of laser channel should be used for image acquisition when staining with a diluted stock solution of Nile red in PBS, considering that Nile red may be appropriate for different excitation nm in different solvents?

    1. In regards to fluorescent properties of Nile Red, product 19123, only methanol is used as the solvent. No testing is performed using DMSO as the solvent and then further diluting into PBS. The specification for product 19123 calls for excitation at 530 nm and emission at 630-640 nm. This differs somewhat from the Biological Stain Commission's recommendations, which state that the excitation is 553 nm in methanol and emission is at 636 nm. The BSC also states that heptane can be used as a solvent. For heptane, the excitation is 485 nm, and emission is at 525 nm. As a result, the excitation and emission wavelengths vary depending on the solvent utilized. No data is available on the excitation and emission in DMF, DMSO, Xylene, Acetone, or ethanol. Nile Red has very minimal solubility in water at just 0.02%.

  4. Would incubating samples at 2-8°C, rather than at room temperature protected from light, improve results, and would this lower temperature need an extension of the 24-hour incubation period?

    1. Staining procedures with Nile Red are generally conducted at room temperature; however, some protocols may call for higher temperatures. Incubating at 2-8°C is less typical and could require a longer staining period, as lower temperatures may slow down the reaction. For optimal results, it is advisable to run a comparative test with one specimen at room temperature and another at 2-8°C to determine the best conditions for a specific application.

  5. Is Nile Red, Product 19123, soluble in acetone?

    1. This product has not been tested for solubility in acetone. However, a brief literature search suggests that this material may be dissolved at 1 mg/mL in acetone.
