General description

Tween® 20 es un éster de sorbitol de polioxietileno que pertenece a la familia polisorbato. Es un detergente no iónico que tiene un peso molecular de 1 225 dalton, asumiendo 20 unidades de óxido de etileno, 1 sorbitol, y 1 ácido láurico como el ácido graso primario. Las subunidades de óxido de etileno son responsables de la naturaleza hidrófila del tensioactivo, mientras que las cadenas de hidrocarburos proporcionan el ambiente hidrófobo. El sorbitol forma el anillo principal al que se conectan los polímeros de óxido de etileno.[1]
Tween® 20 es un tensioactivo y detergente no iónico ampliamente aplicado en investigación bioquímica y de ciencias de la vida. Su uso predominante consiste en la solubilización de las proteínas de membrana durante el aislamiento de complejos membrana-proteína. Además, es un componente de varios amortiguadores de lavado, bloqueo, dilución y estabilización. Por otro lado, funciona como reactivo en inmunohistoquímica, abarcando técnicas como inmunoelectrotransferencia (western blot) y ELISA. En estas aplicaciones, Tween® 20 desempeña funciones cruciales, como evitar la unión inespecífica de los anticuerpos, minimizar la tinción de fondo y garantizar la distribución uniforme de los reactivos.

Los tensioactivos, que poseen componentes hidrófilos e hidrófobos, se dividen en dos categorías principales: iónicos, que se ionizan en el agua (aniónicos, catiónicos o anfolíticos), y no iónicos, que no se ionizan. El producto que nos ocupa es un tensioactivo no iónico con diversas aplicaciones, como emulsificación, dispersión, solubilización y detergente. Además, puede actuar como solubilizante de las proteínas de membrana.

Application

Detergente no iónico
El TWEEN 20 es un detergente no iónico muy utilizado en aplicaciones bioquímicas. Se ha utilizado como emulsionante para la preparación de emulsiones de aceite en agua estables. El TWEEN 20 se ha utilizado en la preextracción de membranas para eliminar las proteínas periféricas (utilizado al 2 % para la extracción de las proteínas ligadas a las membranas).
El TWEEN 20 se ha utilizado como bloqueante para inmunoanálisis en membrana a una concentración típica del 0,05 %. El TWEEN 20 puede utilizarse para lisar células de mamífero a una concentración comprendida entre el 0,005 y el 0,5%.
El TWEEN 20 se ha utilizado como componente de:
  • el tampón de lavado y el tampón de bloqueo en la inmunoelectrotransferencia[2]
  • el tampón de bloqueo en la inmunohistoquímica[3]
  • el tampón de lavado en el ELISA (enzimoinmunoanálisis de absorción)[4][5]
  • la mezcla de reacción de la PCR (reacción en cadena de la polimerasa)[6]

Preparation Note

  6. You state 0.06 mM as the CMC for this product. What is the stock concentration (in Molar) for this product? Alternatively what is the % to reach that 0.06 mM / CMC concentration? Thank you

    1. The critical micelle concentration (CMC) reported on the product page is a value reported from the literature. This product is a neat or pure liquid. The base molecular weight of this material is 1,225 daltons and the density is 1.095 g/mL. However, this is based on the assumption of 20 ethylene oxide units, 1 sorbitol, and 1 lauric acid.     1. Long-term stability studies have not been performed, however, providing that the product is stored and handled properly, the expected shelf life is not expected to change. Factors such as improper closure and cross-contamination would certainly impact the overall stability and shelf life.

  9. The data sheet for my lot says this is 50% pure. Does that mean it is 50% tween 20?

    1. The 50% is referring to the concentration of lauric acid which occurs in the highest concentration. Fatty acid constituents of this product are determined by transesterification to yield fatty acid methyl esters, which are identified by gas chromatography. Fatty acids that are typically present in the offered Tween products are not a quantitative measurement. These include myristic, palmitic, and stearic acid, for the most part. While the Tween is considered a 'pure' product, the molecular weight is calculated on the assumption of 20 ethylene oxide units, 1 sorbitol, and 1 lauric acid as the primary fatty acid.
