Specificity

Clone 2F15-L1 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects Thrombospondin type-1 domain-containing protein 7A (THSD7A). It targets an epitope within 20 amino acids from the extracellular domain within the N-terminal half.

Immunogen

KLH-conjugated linear peptide corresponding to 20 amino acids from the N-terminal half of human Thrombospondin type-1 domain-containing protein 7A (THSD7A).

Application

Quality Control Testing

Evaluated by Western Blotting in Human kidney tissue lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected THSD7A in Human kidney tissue lysate.

Tested Applications

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected THSD7A in Human kidney tissue sections.

Immunofluorescence Analysis: A 1:100 dilution from a representative lot detected THSD7A in Human kidney tissue sections.

Affinity Binding Assay: A representative lot of this antibody bound THSD7A peptide with a KD of 9.8 x 10-8 in an affinity binding assay.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected THSD7A in Human umbilical vein endothelial cells (HUVEC).

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Target description

Thrombospondin type-1 domain-containing protein 7A (UniProt: Q9UPZ6; also known as THSD7A) is encoded by the THSD7A (also known as KIAA0960) gene (Gene ID: 221981) in human. THSD7A is a single-pass type I membrane glycoprotein that is synthesized with a signal peptide (aa 1-47), which is subsequently cleaved to produce the mature form that contains an extracellular domain (aa 48-1607), a transmembrane domain (aa 1608-1628), and a short cytoplasmic domain (aa 1629-1657). It contains 19 thrombospondin type 1 repeats, multiple WSXW motifs, and one RGD motif (aa 1568-1570). It is detected on kidney podocytes along the glomerular capillary wall. It plays a role in actin cytoskeleton rearrangement. There also exists a soluble form that promotes endothelial cell migration and filopodia formation during sprouting angiogenesis via a focal adhesion kinase-dependent mechanism. THSD7A is a well-known antigenic target of autoantibodies leading to primary membranous glomerulopathy and accounts for about 10% cases of phospholipase A2 receptor-negative membranous glomerulopathy. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

0.3 mg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
