General description

NR4A2 is encoded by the gene mapped to human chromosome 2q24.1.
Nuclear receptor subfamily 4 group A member 2 (NR4A2) is part of transcriptional regulator subfamily, under the family of nuclear hormone receptors. It is a transcription factor expressed in the brain and T-cells.

Specificity

Anti-NR4A2 specifically recognizes human NR4A2.

Application

Anti-NR4A2 antibody produced in rabbit has also been used in immunohistochemistry (IHC).
Anti-NR4A2 antibody produced in rabbit has been used in western blotting.

Biochem/physiol Actions

Nuclear receptor subfamily 4 group A member 2 (NR4A2) acts as an activator of tyrosine hydroxylase. It is crucial for the process of DNA repair by melanocortin-1 receptor.
Nuclear receptor subfamily 4 group A member 2 (NR4A2) receptors function as transcription factors. NR4A2 binds the NGFI-B (nerve growth factor-induced clone B) response element (NBRE) sequence (AAAGGTCA) in the form of monomer. NR4A2 is induced by multiple extracellular signals, including fatty acids, stress, prostaglandins, growth factors, calcium, inflammatory cytokines, peptide hormones, and neurotransmitters in a cell type specific manner. Cyclic adenosine monophosphate (cAMP) triggers the hepatic expression of NR4A2 in response to glucagon and fasting. Diabetic mice with elevated gluconeogenesis also show high levels of NR4A2. NR4A2 is essential for the differentiation of nigral dopaminergic neurons. Mice lacking Nurr1 failed to generate midbrain dopaminergic neurons. NR4A2 mutations have been associated with disorders related to dopaminergic dysfunction, including Parkinson′s disease, schizophrenia and manic depression.

Physical form

Solution in 0.01 M phos­phate buffered saline, pH 7.4, containing 15 mM sodium azide.

Storage and Stability

For continuous use, store at 2–8 °C for up to one month. For extended storage, freeze in working aliquots. Repeated freezing and thawing is not recommended. Storage in “frost-free” freezers is also not recommended. If slight turbidity occurs upon prolonged storage, clarify the solution by centrifugation before use. Working dilutions should be discarded if not used within 12 hours.
