General description

γ-Actin is mainly present in sarcolemma and to a lesser extent in reticular lattice of normal muscles. Increased levels of γ-actin have been linked to dystrophin loss and subsequent cell signaling and gene expression defects.
Monoclonal Anti-γ-Actin (mouse IgG1 isotype) is derived from the hybridoma 2-2.1.14.17 produced by the fusion of mouse myeloma cells (NS1) and splenocytes from BALB/c mice. Actin is one of the most conserved eukaryotic proteins, it is expressed in mammals and birds as at least six isoforms. Actin isoforms show more than 90% overall sequence homology, but only 50-60% homology in their 18 NH2-terminal residues. Actin constitutes for around 15% of the total protein. It exists in globular and filamentous forms.

Specificity

Monoclonal Anti-γ-Actin recognizes human, bovine, canine, mouse, hamster and chicken γ-actin.

Application

Anti-γ-Actin antibody, Mouse monoclonal has been used in :
  • immunofluorescence
  • enzyme linked immunosorbent assay (ELISA)
  • western blotting
  • immunohistochemistry

Mouse monoclonal Anti-γ-Actin antibody has been used for immunofluorescence assays. The antibody can also be used for ELISA, IHC and western blot assays.
Protein lysates from human Jurkat T-leukemic cells were subjected to western blot and probed using monoclonal mouse anti-actin (AC-40) to ensure equal loading.

Biochem/physiol Actions

The two major cytoskeletal proteins implicated in cell motility are actin and myosin. Actin is significantly involved in the microfilament system. It participates in muscle contraction and gene regulation. Actin and myosin are constituents of many cell types and are involved in a myriad of cellular process including locomotion, secretion, cytoplasmic streaming, phagocytosis and cytokinesis. Increased levels of γ-actin have been linked to dystrophin loss and subsequent cell signaling and gene expression defects.

Physical form

Solution in 0.01 M phosphate buffered saline, pH 7.4, containing 15 mM sodium azide.
