DNA/RNA G-quadruplexes (G4s) are four-stranded nucleic acid structures that fold from numerous guanine (G)-rich sequences found in the genome and transcriptome. Their abundance and localization in critical regulatory regions make them key players in genetic diseases (cancer, neuropathology), but not only (e.g., viral infection). For this reason G4s have garnered intense interest as potential targets in different therapeutic areas.
The BioTracker™ BioCyTASQ G-quadruplex G4 cell probe is optimized for detection (1) and/or the isolation of G4s (2,3). BioCyTASQ utilizes the template-assembled synthetic G-quartets (TASQ) (4) concept as a biomimetic ligand for G-quadruplexes, a design which results in high specificity for folded G4.

Features and Benefits
BioCyTASQ is suited to visualize G4s in cells in a 2-step protocol referred to as pre-targeted G4 imaging, which relies on the use of a fluorescent streptavidin (1). BioCyTASQ is also suited to isolate G4s by affinity capture experiments, in a protocol named G4RP for RNA G4s, (2,3) or G4DP for DNA G4s (5)). These G4-selective chemoprecipitations allow for their identification by either PCR, RT-PCR, or sequencing.
