General description

Ketone bodies, 3-hydroxybutyric acid (BOH) and acetoacetic acid (AcAc), are produced in the liver primarily from oxidation of fatty acids,[1] and are normally present at low concentrations in urine and blood. Increased ketone concentrations in the blood may lead to metabolic acidosis, which has been associated with diabetes, childhood hypoglycemia, growth hormone deficiency, alcohol or salicylate intoxication, and inborn errors of metabolism.[2][3]

Application

Ketone Body Assay has been used to measure the release of ketone bodies in the human liver cancer cell line HepG2 culture medium.[4]

Suitability

Suitable for the detection of ketone bodies (acetoacetic acid and 3-hydroxybutyric acid) in serum, plasma, urine, and other biological samples.

Principle

In this kit, AcAc and BOH levels are determined using an enzymatic assay based on 3-hydroxybutyrate dehydrogenase catalyzed reactions, in which the change in NADH absorbance measured at 340 nm is directly related to either the AcAc and BOH concentrations.

    1. If this product has an expiration or retest date, it will be shown on the Certificate of Analysis (COA, CofA). If there is no retest or expiration date listed on the product's COA, we do not have suitable stability data to determine a shelf life. For these products, the only date on the COA will be the release date; a retest, expiration, or use-by-date will not be displayed.
      For all products, we recommend handling per defined conditions as printed in our product literature and website product descriptions. We recommend that products should be routinely inspected by customers to ensure they perform as expected.
      For products without retest or expiration dates, our standard warranty of 1 year from the date of shipment is applicable.

    1. Products may be shipped at a different temperature than the recommended long-term storage temperature. If the product quality is sensitive to short-term exposure to conditions other than the recommended long-term storage, it will be shipped on wet or dry-ice. If the product quality is NOT affected by short-term exposure to conditions other than the recommended long-term storage, it will be shipped at ambient temperature.     1. Certainly, whole blood or plasma collected in EDTA is appropriate for use with this kit. During the development of the kit, these sample types were tested as they are commonly used by researchers. And also AcAc (Acetoacetic acid) is quite unstable, so it is advised using fresh samples or ones that have been frozen within the month.
