  1. Are adult red blood cells failing to properly elute hemoglobin after incubation in the Citrate Phosphate Buffer working solution? Could the pH of the Citrate Phosphate Buffer be too high? Additionally, is it feasible to increase the amount of agitation to aid in eluting the adult hemoglobin?

    1. To elute adult hemoglobin, pure/undenatured 80% alcohol is necessary. The use of histological or reagent grade alcohol may result in inadequate elution.

  2. Is it advisable to switch to the published procedure, which requires a 5-minute incubation, considering the change in cell appearance after immersing the slides for 2 minutes in the buffer?
