The NHP Cytokine Panel 1 (PRCYTOMAG-40K) kit was modified such that the TNF-a standard lot is value assigned. The first standard lot with this change was SC840-22K. This adjustment should not cause QCs to read out of range as it is still calibrated to the gold standard. The raw material used for the TNF-a standard has not changed. TNFα was identified as an analyte that would benefit from a more robust production process.

General description
The MILLIPLEX® Non-Human Primate Cytokine panel is to be used for the simultaneous quantification of any or all of the following in tissue/cell lysate and culture supernatant samples and serum or plasma samples: G-CSF, GM-CSF, IFNγ, IL-1β, IL-1ra, IL-2, IL-4, IL-5, IL-6, IL8, IL-10, IL-12/23(p40), IL-13, IL-15, IL-17A, MCP-1, MIP-1β, MIP-1α, sD40L, TGF-α, TNF-α, VEGF, and IL-18.
The Luminex® xMAP® platform uses a magnetic bead immunoassay format for ideal speed and sensitivity to quantitate multiple analytes simultaneously, dramatically improving productivity while conserving valuable sample volume.

Has the concentration of the TNFa standard in the NHP Cytokine Milliplex kit changed? It was previously 10,000 pg/ml but is now listed on the QC Analysis Sheet and changes with each lot.

Could you provide information about the specificity and cross-reactivity between IL-1b and IL-18 for the antibodies in kit PRCYTOMAG-40K, which is used for detecting monkey cytokines? There is concern that we may not be able to measure both molecules simultaneously.

Individual standards in the kit were tested using multiplexed beads. It was observed that both IL-1b against IL-18 standards and IL-18 against IL-1b standards showed levels below background.

Does the IL-1B antibodies in the NHP cytokine panel (cat # PRCYTOMAG-40K) measure total IL-1b or the monomeric/bioactive IL-1b?

The IL-1B antibodies included in the NHP cytokine panel (cat # PRCYTOMAG-40K) have not been specifically tested for their ability to measure the active form only. Based on available antibody information, it is known that they can detect both natural and recombinant forms of IL-1B and that they are monoclonal antibodies. However, additional specificity information is not available.

What is the reason for the much lower median readings on the standards in my Milliplex assay after accidentally washing off the detection antibody before adding SAPE?
1 answer-
Lower signal is expected when the detection antibody is washed off and not incubated with the SAPE. SAPE has multiple binding sites, and when incubated with the detection antibodies, it leads to the formation of larger complexes, resulting in an increased signal.
The drop in signal should be equal across the entire plate, and if the quality control results are within the range, the calculated sample results should be acceptable.
