General description

Ceramides comprise sphingoid base attached to fatty acyl chain by an amide bond. They vary in length, saturation and hydroxylation of the fatty acid chain and sphingoid base. Ceramides are synthesized by sphingomyelinase in salvage and de novo synthesis pathway. In brain, ceramide synthases mediate the synthesis of ceramides.

  • in ultra performance liquid chromatography-electrospray ionization-quadrupole time-of-flight mass spectrometry (UPLC-ESI-Q-TOF-MS)
  • in liquid chromatography with a hydrophilic interaction column
  • in liposome preparation to test it effect on human α–synuclein aggregation

Biochem/physiol Actions

Ceramides are signaling molecules which play a key role in cellular events. Ceramides promote β-amyloid synthesis and induce neuronal death. Ceramide levels are associated with pathogenesis of many disease states. Impairment of ceramide metabolism is implicated in Parkinson′s and Alzheimer′s disease. High levels of ceramide is also associated with depression.
