General description

Choline acetyltransferase (ChAT) has a molecular weight of about 68 kDa. It is synthesized in the parakaryon and is transported to the axon terminals. ChAT belongs to the choline/carnitine acyltransferase family. It seems that only cholinergic neurons have significant amounts of ChAT making anti-choline acetyltransferase a useful specific marker. Choline acetyltransferase is found in cholinergic neurons in brain and spinal cord.

Specificity

Expected to cross-react with Avian, chicken, guinea pig, monkey, rat, and opposum.

Immunogen

Human placental enzyme.

Application

Anti-Choline Acetyltransferase Antibody has been used in:
  • immunohistochemical staining
  • western blotting
  • immunocytochemistry

Immunohistochemistry:
A previous lot of this antibody was used at 1:100.

Optimal working dilutions and protocols must be determined by end user.

Biochem/physiol Actions

Choline acetyltransferase (CHAT) is involved in the synthesis of acetylcholine in neurons. It catalyzes the reversible synthesis of acetylcholine (ACh) from acetyl CoA and choline at cholinergic synapses. CHAT catalyzes the transfer of an acetyl group between acetyl-coenzyme A and choline. Mutations in the gene encoding this protein have been associated with congenital neuromuscular disorders.

Quality

Routinely evaluated by Western Blot on mouse brain lysates.

Western Blot Analysis:
1:1000 dilution of this lot detected CHAT on 10 μg of mouse brain lysates.

Storage and Stability

Maintain at -20°C in undiluted aliquots for up to 1 year from date of receipt. Avoid repeated freeze/thaw cycles.
