General description

Dithiothreitol (DTT) is used in proteomics applications to maintain sulfhydryl (-SH) groups in the reduced state and for quantitative reduction of disulfide (-S-S-) groups. It is a frequently used reagent in buffers due to its ability to reduce the oxidation state of a protein sample and preserves enzymatic activity. DTT is oxidized to the cyclic disulfide during the reduction of other disulfides in the solution.
Dithiothreitol is an effective reducing agent for protein analysis. The sealed-ampule format is convenient and easy to use when preparing samples for electrophoresis or other applications and avoids waste and loss of potency.

Application

DL-Dithiothreitol solution has been used:
  • in western blot analysis
  • as a supplement in lysis buffer to study its effects on sperm lysis
  • as a component of cold homogenization buffer, and chilled wash buffer for the isolation of nuclei from frozen rat tissues
  • in sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE), chromatography, modification of cysteine-containing compounds

Features and Benefits

Low toxicity and reduced odor compared to other thiol compounds.

Questions

1–8 of 8 Questions
  1. Is the DTT supplied in a glass ampule?

    1 answer
    1. Yes, this product is packaged in a 2 mL clear glass sealed ampule.

      Helpful?

  2. How can I determine the shelf life / expiration / retest date of this product?

    1 answer
    1. If this product has an expiration or retest date, it will be shown on the Certificate of Analysis (COA, CofA). If there is no retest or expiration date listed on the product's COA, we do not have suitable stability data to determine a shelf life. For these products, the only date on the COA will be the release date; a retest, expiration, or use-by-date will not be displayed.
For all products, we recommend handling per defined conditions as printed in our product literature and website product descriptions. We recommend that products should be routinely inspected by customers to ensure they perform as expected.
For products without retest or expiration dates, our standard warranty of 1 year from the date of shipment is applicable.
For more information, please refer to the Product Dating Information document: https://www.sigmaaldrich.com/deepweb/assets/sigmaaldrich/marketing/global/documents/418/501/product-dating-information-06-25-mk.pdf

  3. What is the ideal storage temperature for this product? Most DTT solutions must be stored at -20, but this was shipped at RT.

    1 answer
    1. In the original sealed ampule, this product is stable for at least two years at room temperature. Please see the link below to review additional information available in the product datasheet:
      https://www.sigmaaldrich.com/deepweb/assets/sigmaaldrich/product/documents/629/609/646563dat.pdf

      Helpful?

  4. How is shipping temperature determined? And how is it related to the product storage temperature?

    1 answer
    1. Products may be shipped at a different temperature than the recommended long-term storage temperature. If the product quality is sensitive to short-term exposure to conditions other than the recommended long-term storage, it will be shipped on wet or dry-ice. If the product quality is NOT affected by short-term exposure to conditions other than the recommended long-term storage, it will be shipped at ambient temperature. As shipping routes are configured for minimum transit times, shipping at ambient temperature helps control shipping costs for our customers. For more information, please refer to the Storage and Transport Conditions document: https://www.sigmaaldrich.com/deepweb/assets/sigmaaldrich/marketing/global/documents/316/622/storage-transport-conditions-mk.pdf

  5. after opening the ampule is DTT still usable or only single use?

    1 answer
    1. This product is intended for single use. The stability of the solution after opening the ampule has not been determined. Please see the link below to review the product datasheet:
      https://www.sigmaaldrich.com/deepweb/assets/sigmaaldrich/product/documents/629/609/646563dat.pdf

  6. how to use DDT

    1 answer
    1. 646563 (DL-Dithiothreitol solution, DTT) could be applied for research, such as western blot analysis, lysis buffer, etc. The procedure of SDS-PAGE sample preparation with DTT has been listed in the datasheet: https://www.sigmaaldrich.com/deepweb/assets/sigmaaldrich/product/documents/629/609/646563dat.pdf
For other applications, please check the "Peer Reviewed Papers" section on the web for more literature: https://www.sigmaaldrich.com/product/sigma/646563

  7. What is the expiration time for this product?

    1 answer
    1. This product is not assigned an expiration date or recommended retest date. Products with no expiration date or recommended retest date should be routinely inspected by customers to ensure they perform as expected. These products are also subject to a one year warranty from the date of shipment.
