General description

Amyloid Beta refers to the short peptides that are released into circulation following the proteolysis of the transmembrane protein amyloid precursor protein (APP), whose function is unclear but thought to be involved in neuronal development and function. The most common peptides generated from APP cleavage are Aβ1-40 and Aβ1-42, with the latter isoform implicated in the formation of toxic oligomers, fibrils, and plaques in the diseased brain. Reduced levels of soluble Aβ1-42 in the cerebrospinal fluid (CSF) are associated with Alzheimer’s disease (AD), leading to the adoption of this peptide as an AD biomarker. The presence of AB40, AB42 in patient CSF and plasma changes as an individual ages, and increases in AB42 levels, and more importantly, the AB42/AB40 ratio, are recognized as correlating with dementia and the onset of Alzheimer’s.
The SMC® Amyloid Beta 1-40 High Sensitivity Immunoassay Kit contains all reagents required to perform the assay. This assay takes advantage of a standard immunoassay workflow configured in a standard plate format. Assay protocols are similar to existing sandwich ELISA methods with two key differences: 1) Elution buffer disrupts the sandwich, separating the labeled detection antibody for quantification. 2) The Erenna and SMCxPRO immunoassay systems detect analytes using Single Molecule Counting (SMC) technology.
