Spécificité

Clone 2K11 is ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects PTPN21. It targets an epitope within 23 amino acids from the C-terminal half.

Immunogène

KLH-conjugated linear peptide corresponding to 23 amino acids from the C-terminal half of human Tyrosine-protein phosphatase non-receptor type 21 (PTPN21).

Application

Quality Control Testing

Evaluated by Western Blotting in ACHN cell lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected PTPN21 in ACHN cell lysate.

Tested applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected PTPN21 in A549, PC12, and Raw264.7 cell lysates.

Affinity Binding Assay: A representative lot of this antibody bound Ptpn21 peptide with a KD of 1.3 x 10-6 in an affinity binding assay.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected PTPN21 in human gallbladder tissue sections.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected PTPN21 in A549 cella.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user

Description de la cible

Tyrosine-protein phosphatase non-receptor type 21 (UniProt: Q16825; also known as EC:3.1.3.48, Protein-tyrosine phosphatase D1) is encoded by the PTPN21 (also known as PTPD1) gene (Gene ID: 11088) in human. PTPN21 is a member of the non-receptor protein-tyrosine phosphatase family. Its tyrosine phosphatase domain is localized to amino acids 896-1167. It is mainly localized in the cytoplasm and is phosphorylated by Src kinase. It also upregulates Src by dephosphorylating it at Tyr 527 in response to stimulation by growth factors. PTPN21 is reported to form a complex with actin, Src, and focal adhesion kinase and localizes at focal adhesion sites. It exerts major effects on cell adhesion, scattering, and cell migration. It is reported to specially interact with ETK and its residues 726-848 are essential for this interaction. ETK-PTPN21 interaction stimulates ETK kinase activity that results in STAT3 activation. PTPN21 is shown to directly dephosphorylate ErbB1 and ErbB4, which results in upregulation of its downstream signaling. It results in the constitutive activation of Src and increase in the activity of Elk-1. PTPN21 is reported to negatively regulate ICAM-1 expression in human keratinocytes. PTPN21 is overexpressed in human gastric B-cells non-Hodgkin s lymphoma tissue where it promotes STAT5 activity. It also exhibits the highest mutation frequency in certain colon cancer with microsatellite instability. This ZooMAbZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Forme physique

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Stockage et stabilité

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
