Spécificité

Clone 1F22 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects Early growth response protein 1 (EGR1). It targets an epitope within 23 amino acids from the N-terminal half.

Immunogène

KLH-conjugated linear peptide corresponding to 23 amino acids from the N-terminal half of human Early growth response protein 1 (EGR1).

Application

Quality Control Testing

Evaluated by Western Blotting in HeLa cell lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected EGR1 in HeLa cell lysate.

Tested applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected EGR1 in PC3 cell lysate.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected EGR1 in human prostate tissue sections.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected EGR1 in PC3 cells.

Affinity Binding Assay: A representative lot of this antibody bound EGR1 with a KD of 1.3 x 10-8 in an affinity binding assay.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Early growth response protein 1 (UniProt: P18146; also known as EGR-1, AT225, Nerve growth factor-induced protein A, NGFI-A, Transcription factor ETR103, Transcription factor Zif268, Zinc finger protein 225, Zinc finger protein Krox-24) is encoded by the EGR1 (also known as KROX24, ZNF225) gene (Gene ID: 1958) in human. EGR1 is a member of the EGR C2H2-type zinc-finger protein family that serves as a transcriptional regulator. It recognizes and binds to the DNA sequence 5′-GCG(T/G)GGGCG-3′(EGR-site) in the promoter region of target genes and regulates their transcription. It contains three C2H2-type Zinc fingers (a 338-362, 368-390, 396-418). The first, most N-terminal zinc finger binds to the 3′-GCG motif, the middle zinc finger interacts with the central TGG motif, and the C-terminal zinc finger binds to the 5′-GCG motif. It is shown to binds double-stranded target DNA, irrespective of the cytosine methylation status. It displays reduced affinity for target DNA where cytosines have been oxidized to 5-hydroxymethylcytosine and does not bind target DNA where cytosines have been oxidized to 5-formylcytosine or 5-carboxylcytosine. EGR1 plays an important role in regulating the response to growth factors, DNA damage, and ischemia. It is reported to activate the expression of p53/TP53 and TGFB1, and thereby prevents tumor formation and is shown to be essential for normal progress through mitosis and normal proliferation of hepatocytes after partial hepatectomy. It is also regulates the expression of proteins such as IL-1b and CXCL2 that are involved in inflammatory processes and development of tissue damage after ischemia. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Forme physique

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

30 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Stockage et stabilité

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
