General description

Calcium channel, L type α-1 polypeptide isoform 2 (CACNA1D) is expressed in many tissues and possesses six transmembrane segments, along with a loop lining the pore of the channel. The gene encoding CACNA1D is localized on human chromosome 3p14.3.

Biochem/physiol Actions

Calcium channel, L type α-1 polypeptide isoform 2 (CACNA1D) is crucial for the functioning of the brain and in hearing. It gets activated at a fast rate and withstands high inward Ca2+ currents even at threshold voltages. CACNA1D aids in dendritic signals linked to mitochondrial stress in the brain and is important for estrogen-stimulated Ca2+ influx. Studies have shown that in atrial myocytes, the C terminus of the channel translocates to the nucleus and regulates Ca2+-activated K+ channels. CACNA1D was shown to be upregulated in endometrial cancer and its knockdown showed a reduction in cell proliferation in vitro. Mutations in the gene encoding it has been associated with aldosterone-producing adenomas (APAs), where it leads to the formation of small adenomas.
