General description

Nephrin is strongly expressed in renal glomeruli and is a member of the immunoglobulin family of cell adhesion molecules. It is a 1241-residue transmembrane protein. Mutations in the Nephrin gene result in congenital nephrotic syndrome, an autosomalrecessive disorder characterized by massive proteinuria in utero and nephrosis at birth. Renal glomeruli allow normal kidneys to filter plasma so that it is very pure. Nephrin is expressed in the podocyte slit-diaphragm of the renal glomeruli in a manner that suggests that Nephrin molecules homodimerize in an anti-parallel fashion similar to cadherin interactions in adherens junctions. Thus, Nephrin may constitute the entire extracellular structure of the slit-diaphragm.

Immunogen

Nephrin antibody was raised against a 14 amino acid synthetic peptide from near the carboxy terminus of human
Nephrin. The immunogen is located within the last 50 amino acids of Nephrin.

The action of this antibody can be blocked using blocking peptide SBP2265.

  1. Why does the Nephrin from mouse kidney indicate a molecular weight of a little less than 100 kDa on the Western blot shown on the webpage for Product PRS2265, anti-Nephrin antibody produced in rabbit?

    1. Based on the literature, one would expect the Nephrin to show up at a higher molecular weight. Unfortunately, we do not have a good explanation for the mouse Nephrin band showing up at a molecular weight of a little less than 100 kDa. It might be due to a cleavage event, but we have not read of that happening with Nephrin. The antibody may be detecting an alternative splicing product, but again, we have not seen anything like that mentioned in the literature on Nephrin.The sequence used as the antigen for this antibody does not possess homology to any other protein. And the PRS2265 antibody is quite specific for this sequence, based on the results when using a blocking peptide as shown in lane B of the blot: Supplier is confident that the antibody is detecting Nephrin but is not able to be of more help in explaining the lower-than-expected molecular weight.According to the Uniprot link for mouse nephrin, the mature form of mouse Nephrin has been described as a protein that is a single polypeptide of 1221 amino acids (1256 minus the signal peptide of 35 amino acids)So based on the amino acids alone, one would expect a band close to 130 kDa.
