General description

Tryptic Soy Broth (TSB) is a general-purpose culture medium designed for the cultivation and enrichment of a wide range of microorganisms, including fastidious and non-fastidious bacteria, fungi, and yeasts. Widely used in microbiological testing, sterility testing, and microbial enumeration, TSB is formulated to support robust microbial growth. The medium contains a combination of casein and soya peptones, which provide essential nutrients such as organic nitrogen, amino acids, and peptides. The peptones in this medium offer enhanced growth-promoting properties. Sodium chloride maintains osmotic balance, while glucose and natural sugars from soya peptone serve as fermentable carbohydrate sources. Dipotassium hydrogen phosphate acts as a buffer to stabilize the pH. Tryptic Soy Broth is particularly effective for isolating organisms from blood or other body fluids. It is often used in the tube dilution method for determining antibiotic susceptibility. The addition of a small amount of agar (approximately 0.05–0.2%) renders the broth suitable for cultivating obligate anaerobes, such as Clostridium species. Anticoagulants, such as sodium polyanetholesulfonate or sodium citrate, may be added before sterilization to enhance their utility in blood culture applications. For blood cultures, 5–10 mL of blood can be added to 50 mL of medium. This nutrient-rich medium is an essential tool for a variety of microbiological applications, ensuring reliable and consistent results.

Preparation Note

Suspend 30 g of dehydrated media in 1 litre of purified filtered water. Heat with frequent agitation and boil for one minute. Sterilize at 121°C for 15 minutes. Cool to 45-50°C. Mix gently and dispense into sterile Petri dishes or sterile culture tubes.
