Application

Trifluoroacetic acid (TFA) can be used as a solvent in a reverse-phase HPLC system to separate peptides because it enhances peak resolution. It also has low absorption detection wavelengths and is relatively volatile and easily removed. For the majority of peptide separations, TFA has been used at concentrations ranging from 0.05 to 0.1%(6.5-13 mM).

Trifluoroacetic acid (TFA) remains the dominant mobile phase additive for reversed-phase high-performance liquid chromatography (RP-HPLC) of peptides after more than two decades since its introduction to this field. Generally, TFA has been employed in a concentration range of 0.05-0.1% (6.5-13
