General description

Mammalian HADC1 (also designated HD1), HDAC2 (also designated mammalian RPD3) and HDAC3, all are related to the yeast transcriptional regulator Rpd3p, are identified as histone deacetylases. The deacetylation of histones is associated with transcriptional silencing.

Specificity

Oher species cross-reactivity not tested.
Recognizes and is specific for HDAC1, Mr 65 kDa. May detects a non-specific protein at ~35 kDa.

Immunogen

6X-His-tagged fragment representing amino acids 53-482 of mouse histone deacetylase 1 (HDAC1).

Application

Immunoprecipitation:
5 μg of a previous lot immunoprecipitated HDAC1 from 500 μg of 3T3/A31 RIPA lysate.

Immunocytochemistry:
This antibody has been reported to immunostain HDAC1 in 1% paraformaldehyde fixed, 0.1% Triton X-100 permeabilized 3T3 cells.

HDAC Activity:
This antibody has been reported to immunoprecipitate active HDAC.

Purified rabbit polyclonal IgG in buffer containing 0.1 M Tris-glycine, pH 7.4, 0.15 M NaCl, containing 0.05% sodium azide. Frozen at -20ºC.

Storage and Stability

Stable for 1 year at -20°C from date of receipt.

Handling Recommendations:
Upon first thaw, and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance.

Analysis Note

Control
Positive Antigen Control: Catalog #12-305, 3T3/A31 lysate. Add 2.5 μL of 2-mercapto-ethanol/100 μL of lysate and boil for 5 minutes to reduce the preparation. Load 20 μg of reduced lysate per lane for minigels.
