Specificity

Clone 1K9 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects Rictor. It targets an epitope within 22 amino acids from the C-terminal region.

Immunogen

KLH-conjugated linear peptide corresponding to 22 amino acids from the C-terminal region of human Rictor.

Application

Quality Control Testing

Evaluated by Western Blotting in HEK293 cell lysate.

Western Blotting Analysis (WB): A 1:1,000 dilution of this antibody detected Rictor in HEK293 cell lysate.

Tested applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected Rictor in MDA-MB-231 cell lysate.

Affinity Binding Assay: A representative lot of this antibody bound Rictor with a KD of 1.1 x 10-6 in an affinity binding assay.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected Rictor in human colon tissue sections.

Target description

Rapamycin-insensitive companion of mTOR (UniProt: Q6R327; also known as AVO3 homolog, hAVO3, Rictor) is encoded by the RICTOR (also known as KIAA1999) gene (Gene ID: 253260) in human. The mammalian target of rapamycin (mTOR) is an evolutionarily conserved serine/threonine kinase that acts as a nutrient sensor and regulates growth factor signal transduction and cellular metabolism. It is an important regulator of protein synthesis, translation initiation, transcription of ribosomal proteins, and the synthesis of rRNA and tRNA. Cellular mTOR is composed of two large protein complexes, known as mTORC1 and mTORC2. The mTORC1 complex is sensitive to rapamycin and consists of mTOR, Raptor, and mLST8 and two negative regulators, PRAS40 and DEPTOR. The Raptor unit serves as a scaffolding protein to bridge the interaction between mTOR and its substrates. The mTORC2 complex that is insensitive to rapamycin, contains mTOR, Rictor, Sin1, and mLST8, Protor, Hsp70, and DEPTOR. This complex plays a role in actin cytoskeleton organization and phosphorylates Akt on Ser473 to fully activate it. Rictor is shown to be essential for Akt phosphorylation at Ser 473. Insensitivity of mTORC2 to rapamycin is reported to be due to the presence of Rictor. Rictor can be phosphorylated at Thr 1135 by ribosomal protein S6 kinase B1 (RPS6KB1), which leads to inhibition of mTORC2 and Akt1 signal transduction. Alterations in RICTOR gene have been identified in breast, pancreatic, colon, and lung cancers. Rictor is also involved in tumor progression by regulating the tumor microenvironment through angiogenesis and remodeling of the stroma. In pancreatic tumors, blockage of Rictor is shown to inhibit HIF-1 expression and reduce the level of VEGF-A. This ZooMAbZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

30 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
