General description

Placental growth factor (PLGF) exists in two isoforms namely PLGF1 and PLGF2. This protein is a homodimeric glycoprotein with molecular weight of 46–50kDa and is a member of the VEGF (vascular endothelial growth factor) subfamily. PLGF is encoded by the gene mapped to human chromosome 14q24-q31 regions. PLGF is expressed mainly in the placental trophoblast during pregnancy and is also present in various other tissues such as the villous trophoblast.

Biochem/physiol Actions

PLGF might enhance the production of peripheral blood mononuclear cell-vascular endothelial growth factor (PBMC-VEGF). Consequently, PlGF might facilitate VEGF production in the inflammatory joint. Soluble fms related tyrosine kinase 1 (Flt-1), which is considered to be an anti-angiogenic factor interacts with and inhibit the activity of PLGF. Therefore, PLGF level is lowered and the Flt-1 level is increased in placental tissue of women with pre-eclampsia (PE). PLGF is associated with the pathogenesis of rheumatoid arthritis (RA) and various other arthropathies. PLGF acts as a ligand for VEGFR-1 and is implicated in various physiological functions such as tumor angiogenesis, wound healing and hematopoietic progenitors recruitment.

Physical form

Lyophilized from 0.22 μm filtered solution in PBS. Generally 5-8% Mannitol or trehalose is added as a protectant before lyophilization.

Reconstitution

Centrifuge the vial prior to opening. Reconstitute in sterile PBS, pH 7.4 to a concentration of 50 μg/mL. Do not vortex. This solution can be stored at 2-8°C for up to 1 month. For extended storage, it is recommended to store at -20°C.
