General description

The pacFA lipid contains a photoactivable diazirine ring with a clickable alkyne group.

Biochem/physiol Actions

D-galactosyl-β-1,1′ N-(9-(3-pent-4-ynyl-3-H-diazirin-3-yl)-nonanoyl)-D-erythro-sphingosine (pacFA GalCer) may be used for a global analysis of protein-lipid interactions in living cells. pacFA acts as a precursor for the biosynthesis of bifunctional lipids. Proteins in contact with the bifunctional lipids are then cross-linked by ultraviolet (UV) irradiation of the diazirine ring. Finally, click chemistry is used to label the alkyne with a reporter molecule.
