Useful synthetic building blocks for various chemical biology applications requiring a bioorthogonal alkyne. Ben Cravatt′s lab recently developed clickable, nucleophilic hydrazine probes for "reverse-polarity" activity-based protein profiling, or (RP)-ABPP. This propargyl hydrazine was used as an alkyl probe to capture protein-bound electrophiles in cells where the hydrazine acts as the nucleophile and alkyne enables protein enrichment and detection. Such methods facilitate discovery of protein modifcations that may not be predicted through typical protein sequence analyses.
