Spécificité

Clone 1E14 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects GPR37. It targets an epitope within 19 amino acids from the first extracellular domain within the N-terminal half.

Application

Quality Control Testing

Evaluated by Western Blotting in THP-1 cell lysate.

Western Blotting Analysis: A 1:10,000 dilution of this antibody detected GPR37 in THP-1 cell lysate.

Tested Applications

Western Blotting Analysis: A 1:10,000 dilution from a representative lot detected GPR37 in lysate from SH-SY5Y cells and 1:1,000 dilution in lysate from Human brain tissue and Rat brain tissue..

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected GPR37 in Human cerebellum tissue sections.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected GPR37 in SH-SY5Y cells.

Affinity Binding Assay: A representative lot of this antibody bound GPR37 peptide with a KD of 4.7 x 10-8 in an affinity binding assay.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Description de la cible

Prosaposin receptor GPR37 (UniProt: O15354; also known as Endothelin B receptor-like protein 1, ETBR-LP-1, G-protein coupled receptor 37, Parkin-associated endothelin receptor-like receptor, PAEL-R, PAELR) is encoded by the GPR37 gene (Gene ID: 2861) in human. GPR37 is an orphan G-protein coupled receptor that is exclusively expressed in the nervous system. It is a multi-pass membrane protein with seven transmembrane domains and is synthesized with a signal peptide (aa 1-26) that is subsequently cleaved off to produce the mature protein. The mature GPR37 is exported from the endoplasmic reticulum to the cell surface, where its long extracellular N-terminus is rapidly and constitutively cleaved by metalloproteinases. It serves as a receptor for the neuroprotective and glioprotective factor prosaposin Ligand binding is shown to induce its endocytosis, followed by an ERK phosphorylation cascade. It is reported to mediate the control of oligodendrocyte differentiation. GPR37 expression and mRNA expression are significantly increased in substantia nigra of sporadic Parkinson s disease cases. The level of ecto-GPR37 peptides is also significantly increased in the cerebrospinal fluid of Parkinson s disease subjects, but not in Alzheimer s disease. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Forme physique

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Stockage et stabilité

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
