Clone 1C11is a ZooMAb® recombinant rabbit monoclonal antibody that specifically detects PKD/PKC . It targets an epitope within 18 amino acids from the C-terminal region of the protein.

Application

Quality Control Testing

Evaluated by Western Blotting in lysate from LNCaP cells.

Western Blotting Analysis (WB): A 1:10,000 dilution of this antibody detected PKD/PKC in lysate from LNCaP cells.

Tested Applications

Western Blotting Analysis: A 1:10,000 dilution from a representative lot detected PKD/PKC in lysate from HuH-7 cells.

Affinity Binding Assay: A representative lot of this antibody bound with PKD/PKC peptide with at least ten thousand-fold (10,000X) higher than with non-specific control peptide.

Immunocytochemistry Analysis: A 1:1,000 dilution from a representative lot detected PKD/PKC in LNCaP cells.

Immunohistochemistry (Paraffin) Analysis: A 1:1,000 dilution from a representative lot detected PKD/PKC in human cerebral cortex tissue sections.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Description de la cible

Serine/threonine-protein kinase D1 (UniProt: Q15139; also known as Protein kinase C mu type, EC:2.7.11.13, Protein kinase D, nPKC-D1, nPKC-mu) is encoded by the PRKD1 (also known as PKD, PKD1, PRKCM ) gene (Gene ID: 5587) in human. The PRKD1 gene promoter has a CpG island covering 1.2 kb and It includes, the transcription start site and the exon 1. The Serine/threonine-protein kinase D1 protein structure features a conserved catalytic Protein Kinase domain at the C-terminus and, an N-terminal regulatory domain that includes a diacylglycerol (DAG)/phorbol ester-binding C1 domain and a pleckstrin homology (PH) domain. Typically, the protein is found in the cytoplasm; however, upon activation, it can translocate to the plasma membrane, golgi appartaus or mitochondria. This translocation is crucial for targeting specific substrates and implies a spatial regulation of its kinase activity. PRKD1 is ubiquitously expressed in various human tissues, with significant expression in the heart, skeletal muscle, and brain. It is also expressed in cells of the normal mammary gland. The protein is involved in many cellular processes, including Golgi body membrane integrity and transport, cell migration and differentiation, MAPK8/JNK1 and Ras pathway signaling, MAPK1/3 (ERK1/2) pathway signaling, vesicle transport, cell survival, and regulation of cell shape and adhesion. In mammary glands, it preserves the epithelial phenotype by inhibiting epithelial-to-mesenchymal transition. Overexpression and dysregulation of PRKD1 are observed in several cancers, including prostate, gastric, and pancreatic cancers, suggesting a role in tumor progression and metastasis. Moreover, PRKD1 has been linked to cardiovascular diseases and neurodegenerative disorders, where its signaling pathways impact cell survival and function. There is also emerging evidence suggesting PRKD1′s role in immune system modulation, potentially impacting autoimmune diseases and inflammation. This ZooMAb® recombinant monoclonal antibody, generated by our proprietary technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Forme physique

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Stockage et stabilité

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
