Spécificité

Clone 1B2 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects Pyruvate Kinase PKM. It targets an epitope within 15 amino acids from the C-terminal region.

Immunogène

KLH-conjugated linear peptide corresponding to 15 amino acids from the C-terminal region of human Pyruvate kinase PKM.

Application

Quality Control Testing

Evaluated by Western Blotting in NIH3T3 cell lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected PKM2 in NIH3T3 cell lysate.

Tested applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected PKM2 in lysates from C6, A549, and MCF7 cells and Human Small Intestine tissue.

Immunohistochemistry (Paraffin) Analysis: A 1:1,000 dilution from a representative lot detected PKM2 in human testis tissue sections.

Affinity Binding Assay: A representative lot of this antibody bound PKM2 with a KD of 2.4 x 10-8 in an affinity binding assay.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected PKM2 in MCF7 and C6 cells.

Pyruvate kinase PKM (UniProt: P14618; also known as EC:2.7.1.40, Cytosolic thyroid hormone-binding protein, CTHBP, Opa-interacting protein 3, OIP-3, Pyruvate kinase 2/3, Pyruvate kinase muscle isozyme, Thyroid hormone-binding protein 1, THBP1, Tumor M2-PK, p58) is encoded by the PKM (also known as OIP3, PK2, PK3, PKM2) gene (Gene ID: 5315) in human. PKM is a glycolytic enzyme that catalyzes the transfer of a phosphoryl group from phosphoenolpyruvate (PEP) to ADP, generating ATP. Four different isozymes of pyruvate kinase have been described in mammals (L, R, M1, and M2). M1 is the main isozyme found in muscle, heart, and brain whereas M2 isozyme is found in early fetal tissues as well as in most cancer cells. PKM2 can exist as a monomer and homotetramer. It exists as a monomer in the absence of D-fructose 1,6-bisphosphate (FBP), and reversibly associates to form a homotetramer in the presence of FBP. It is allosterically activated by FBP. The ratio between the highly active tetrameric form and nearly inactive dimeric form determines whether glucose carbons are channeled to biosynthetic processes or used for glycolytic ATP production. PKM2 is shown to translocate to the nucleus in response to different apoptotic stimuli and nuclear translocation is sufficient to induce cell death that is caspase independent and is not dependent of its enzymatic activity. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Forme physique

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Stockage et stabilité

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
