Description générale

Anti-phospho-AKT (pThr450) antibody, mouse monoclonal (mouse IgG2a isotype) is derived from the AK-11 hybridoma produced by the fusion of mouse myeloma cells and splenocytes from an immunized BALB/C mouse. Serine/threonine kinase (AKT) also known as protein kinase B (PKB) belongs to the family of serine/threonine kinases. It consists of three isoforms such as AKT1, AKT2 and AKT3. AKT1 gene is located on the human chromosome at 14q32.33.

Spécificité

Anti-phospho-AKT (pThr450) antibody, Mouse monoclonal specifically recognizes all three AKT isoforms phosphorylated at Thr450 (AKT1, AKT2 and AKT3).

Immunogène

Synthetic peptide corresponding to human AKT1 pThr450

Application

Anti-phospho-AKT (pThr450) antibody, mouse monoclonal may be used in:
  • immunoblotting
  • immunofluorescence
  • immunoprecipitation

Actions biochimiques/physiologiques

Serine/threonine kinase (AKT) plays an important role in the control of cell cycle and cell proliferation. It is also involved in cell differentiation and apoptosis signaling pathways. AKT is rapidly activated in response to cell stimulation by several growth factors, such as insulin, peroxyvanadate or cellular stresses (such as heat shock). Following activation, the phosphorylated AKT is relocated to the nucleus. Lack of phosphorylation at Thr450 enhances AKT ubiquitination and degradation.

Forme physique

Solution in 0.01 M phosphate buffered saline, pH 7.4, containing 15 mM sodium azide.

Stockage et stabilité

For continuous use, store at 2–8 °C for up to one month. For extended storage, freeze in working aliquots. Repeated freezing and thawing is not recommended. If slight turbidity occurs upon prolonged storage, clarify the solution by centrifugation before use. Working dilution samples should be discarded if not used within 12 hours.
