Description générale

The Human BDNF ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of human BDNF in serum, plasma, cell culture supernatants and urine.
The brain derived neurotrophic factor (BDNF) gene is mapped to human chromosome 11p14.1.[1] BDNF is a member of the neurotrophin family of growth factors.[2] The gene encodes a precursor protein, proBDNF. Mature BDNF (mBDNF) is synthesized by post-translational cleavage of proBDNF. Both proBDNF and mBDNF play crucial roles in cellular signaling.[3]

Application

For research use only. Not for use in diagnostic procedures.
Human BDNF ELISA kit has been used to determine the effect of bone marrow mesenchymal stem cell (MSC) transplantation on BDNF protein expression in the striatum of Tourette syndrome (TS) rats.[4] It has also been used to determine the plasma BDNF levels in GD1 (Gaucher′s disease type 1) patients and control population.[5]
Human BDNF ELISA Kit has been used:
  • to measure the brain-derived neurotrophic factor (BDNF) protein expression[6]
  • to determine BDNF level in the slice culture medium and the extracts of organotypic brain slice cultures (OBSCs)[7]
  • to determine BDNF level in plasma[8]

Actions biochimiques/physiologiques

Brain derived neurotrophic factor (BDNF) is involved in hippocampal function and verbal episodic memory in humans. Hence, variation in the BDNF gene expression alters these neurological functions.[9] BDNF also plays a vital role in vascular function and participates in angiogenesis via the specific receptor tropomyosin-related kinase B (TrkB).[10] It is involved in the pathogenesis of Alzheimer′s disease. ProBDNF interacts with p75 neurotrophin receptor, leading to long-term depression in the hippocampus.[3]

Questions

  1. Could you please confirm if this kit is capable of recognizing both free and bound BDNF?

    1 réponse
    1. Epitope mapping has not been conducted, so the exact binding site is unknown. However, the immunogen is located at aa128-247 (Accession# P23560).
