Description générale

LETM1 (Leucine zipper-EF-hand containing transmembrane protein 1) is a mitochondrial inner membrane protein, and is homologous to yeast protein Mdm38p.[1] It was originally recognized as one of the genes deleted in Wolf-Hirschhorn syndrome. It is a transmembrane protein, which has 14-3-3-like domain in its soluble region, two EF hand Ca²⁺-binding motifs and two coiled-coil domains. The C-terminal of this protein is hydrophilic and faces the matrix, whereas the N-terminal is hydrophobic and spans the membrane. It has a molecular weight of 83.4kDa. This gene is located on human chromosome 4p16.3.

Actions biochimiques/physiologiques

LETM1 (Leucine zipper-EF-hand containing transmembrane protein 1) is responsible for maintaining the shape and volume of mitochondria. It plays a part in mitochondrial translation machinery and mitochondrial biogenesis.[1] It regulates the buffering of mitochondria by controlling Ca²⁺/H⁺ antiporter, and also regulates K⁺/H⁺ ion exchange. Inactivation of this gene is associated with Wolf-Hirschhorn syndrome, where it leads to aberration in mitochondrial functionality. It is responsible for growth and motor delay, as well as seizures which characterize Wolf-Hirschhorn syndrome.[2] It is up-regulated in various human cancers, and is involved in the tumorigenesis of head and neck squamous cell carcinoma (HNSCC). It also predicts poor prognosis in HNSCC.[1] Up-regulation of this gene leads to suppressed mitochondrial biogenesis and ATP synthesis, leading to necrotic cell death.
