Application
Le sérum AB humain est utilisé dans les applications d'ingénierie tissulaire, de transplantation et de thérapie cellulaire pour la croissance de cellules souches mésenchymateuses (CSM) issues de tissu adipeux ou de cellules stromales mésenchymateuses issues de moelle osseuse humaine ; pour la production et la transplantation de greffons de cellules souches épithéliales limbiques standardisées ; pour la croissance ex vivo de cellules NK issues de sang périphérique dans du Milieu de croissance pour cellules souches hématopoïétiques et pour améliorer la technologie de culture des cellules insulaires humaines avant transplantation.
Le sérum humain a été utilisé : dans une solution saline tamponnée au phosphate avec du détergent Tween® (PBST) pour un essai immuno-enzymatique (ELISA) du type "bridging", en guise de simulation d'un échantillon biologique pour évaluer les performances d'un test ELISA sur papier avec des échantillons biologiques complexes, comme diluant pour créer une série de dilutions d'un échantillon de référence afin de générer une courbe d'étalonnage pour du sérum enrichi en NISTmAb, ainsi que pour un échantillon de patient MM2232 atteint de myélome multiple (MM).
Actions biochimiques/physiologiques
Le sérum AB humain, connu sous le nom de sérum OTC (off-the-clot) ou sérum de caillot, est généralement obtenu en laissant coaguler naturellement du sang total. On privilégie le sérum humain obtenu à partir de sang total coagulé car il contient habituellement des facteurs de croissance plus variés et en plus grande quantité, pour une croissance optimale des cellules. Le sérum humain issu de donneurs du groupe sanguin AB ne contient pas d'anticorps dirigés contre les antigènes des groupes A et B. C'est donc un sérum de donneur universel qui convient à différents types de cultures cellulaires comme les lignées de cellules souches humaines.
Conditionnement
Le conditionnement peut varier d'un lot à l'autre en raison d'une pénurie touchant les flacons servant à emballer ce produit. La présence ou l'absence de film rétractable autour du bouchon du flacon n'a aucune incidence sur l’intégrité ou la stérilité du produit.
Avertissement
Chaque donneur est soumis à un test et doit être non-réactif aux anticorps des hépatites B et C et du virus de l'immunodéficience humaine (VIH) par ELISA. Néanmoins, les produits d'origine humaine doivent être considérés comme potentiellement infectieux et traité en conséquence.

Il n'est pas anormal pour nos produits à base de sérum humain d'être troubles. Ce produit est issu d'un mélange de sérum et sa turbidité indique que des lipides sont présents dans le sérum. Aucun produit à base de sérum humain limpide et sans lipide n'est disponible.

Each donor is tested for and found non-reactive to Hepatitis B & C and non-reactive to Human Immunodeficiency Virus (HIV) antibody by ELISA. HSV is not on the list of agents that are tested. If this is "serum" derived from "plasma" the plasma would have contained anti-coagulant. To turn the plasma into serum it has to be clotted. Has this product been clotted to remove the fibrin? Or is it still in fact plasma?

The product has been clotted. Blood is collected and treated with citrate (anticoagulant) which inhibits clotting initially. The blood is then centrifuged. When the spin is complete, layers will appear which allow a differentiation between the cells and the plasma. The plasma is removed and calcium is added to activate the clotting cascade. The preparation is then centrifuged again and the result is a large clot which is pelleted to the bottom of the tube with a serum layer on top.

The use of human serum for a nucleic acid stability assay has not been performed as it is not part of our lab's product characterization focus. However, the citation titled "Improving gene silencing of siRNAs via tricyclo-DNA modification, PMID: 21687522" suggest that product H4522 may be suitable for this application. Human AB serum is added as a supplement to culture media at a concentration of 2-10% to provide a mixture of nutritional, hormonal, growth, and attachment factors. According to the supplier:
Each donor unit used in the preparation of this material was tested and found negative for HBsAG, HBV-NAT, HIV-1/2, HCV, HCV-NAT, HIV-1 NAT, and Syphilis per FDA regulations. The human plasma was collected from male donors' blood type AB in FDA licensed commercial donor establishments located in the USA in accordance with ethical standards. Each donor signed an informed consent form prior to collection, which is traceable by the unique identifiable number to maintain anonymity. A quality assurance system is in place to ensure traceability to individual donor. Documentation to this effect is available upon request.Utile ?

Could you please confirm if the serum contains normal levels of human complement components?

This product has not been treated to remove or inactivate complement components, however no testing has been performed to confirm these levels. It is a pooled product derived from 50 - 70 donors per lot. Any active complement be expected to meet 'normal levels.
