Description générale

Hemoglobin is the major component of red blood cells, and is responsible for their red color. Its normal concentration in erythrocytes is 34%. Hemoglobin is the most important respiratory protein of vertebrates by virtue of its ability to transport oxygen from the lungs to body tissues, and to facilitate the return transport of carbon dioxide.
has not been tested for functional equivalence against native preparations (unlyophilized ferrous hemoglobins).

Application

Hemoglobin A0, ferrous stabilized human has been used:
  • as the non-fluorescent absorber in the preparation of tissue-mimicking phantoms[1]
  • to see how coronary intraplaque hemorrhage evokes a novel atheroprotective macrophage phenotype[2]
  • to study the interference of hemoglobin (Hb) A0, HbA2, and HbS in the assay of HbF[3]

Hemoglobin A0 was used to see how coronary intraplaque hemorrhage evokes a novel atheroprotective macrophage phenotype.[4] It was used in the determination of fetal hemoglobin by time-resolved immunofluorometric assay.[3]

Actions biochimiques/physiologiques

Variation in the haemoglobin (Hb) encoding genes results in haemoglobinopathies such as thalassaemia and atypical structural variants like Hb Lepore and HbE.[5]

Conditionnement

Package size indicates the amount of hemoglobin as determined by the procedure of Drabkin, D.L., J. Biol. Chem., 164, 703 (1946).

Reconstitution

When reconstituted with buffer, gives >85% ferrous hemoglobin.

    1. This product is soluble in water up to 20 mg/mL. Alternatively, a buffer such as phosphate-buffered saline at a neutral pH can be used. Stability in solution has not been tested.

    1. Before lyophilyzation, ficoll & sucrose are added stabilizers & the hemoglobin is dialyzed against 25 mM ammonium bicarbonate containing 0.1 mM EDTA. It is expected that after lyophilization, there should be virtually no or substantially reduced ammonium bicarbonate. Stabilizers are the major components of the final product.

    1. Solutions of ferrous hemoglobin are not stable indefinitely due to the likely autoxidation of the heme group leading to the formation of the ferric derivative (methemoglobin). It is recommended to prepare solutions fresh in deaerated water or buffer.  If necessary, store such prepared solutions no more than 1-2 days at 2-8°C.

    1. Hemoglobin A0 is the major hemoglobin protein of adult hemoglobin. It is a tetrameric protein consisting of 2 alpha and 2 beta chains, each of which contains a heme moiety and iron.

    1. Sigma has not validated this hemoglobin for use in functional studies. The iron in the heme moiety is in the +2 state.  However, this does not mean that this hemoglobin will behave necessarily like native hemoglobin nor be useful for functional studies of the molecule.

    1. Sigma has not evaluated the molecular weight of hemoglobin.  Based on the reported sequence and including the weight of the 4 heme groups and 4 iron atoms, the molecular weight is approximately 64,677 Daltons. The Structure and Action of Proteins, p. 52, Harper and Row (1969).
