Description de la lignée cellulaire

TF-1 have been derived from a patient with erythroleukaemia. They show complete growth dependency on GM-CSF or IL-3, and carry a homogeneous chromosomal abnormality (54,X). Constitutive expression of globin genes indicate commitment of TF-1 to the erythroid lineage, although glycophorin A or carbonyl anhydrase I are not expressed. On incubation with hemin or delta-aminolevulinic acid haemoglobin synthesis was induced, whereas TPA induced differentiation into macrophage-like cells. Erythroprotein also sustains short-term growth, but does not induce differentiation.

Application

TF1 cell line has been used-
  • for tumorigenic studies
  • to study the effect of granulocyte-macrophage colony-stimulating factor (GM-CSF) and interleukin-3 (IL-3) on proliferation and TNFR2 expression in TF1 cells[1]
  • in a cell-based proliferation assay to study the biological activities of anti-IL-6R Nanobody ALX-0061[2]
  • to study the induction of STAT5 phosphorylation by IL-3[3]

Milieu de culture

RPMI 1640 + 2mM Glutamine + 1% Sodium Pyruvate (NaP) + 2-5ng/ml Human Granulocyte Macrophage Colony Stimulating Factor (Human GM-CSF) + 10% Foetal Bovine Serum (FBS). It is recommended to add fresh GM-CSF every 48 hours.

Maintain cultures between 2-9x100,000 cells/ml; 5% CO2; 37°C.

  1. Why is Vapor Phase Liquid Nitrogen preferred for storage of Cell Line cells?

    1. If ampules are immersed into liquid phase of liquid nitrogen, it increases the risk of the liquid seeping into the vial. This could lead to problems of cross-contamination and increased risk of the ampule exploding when thawed.

  2. How many Cell Line cells are in the vial?

    1. Each vial of cells contains 2-3 x 106 cells in 1 ml of freezing media. This is in a 1.8 mL ampule.  For exceptionally large cells, counts may decrease. Suspension cells, generally smaller cells, may contain as many as 4-5 x 106 cells/vial to assure optimal viability upon thaw.

  3. Are the Cell Line cells tested for viral pathogens?

    1. HPA Cultures does not perform any viral testing on the cell lines.

  4. What medium should I use when I thaw the Cell Line cells?

    1. The medium for each cell line is listed on the product page. It is specific to each cell line. If not visible on the Sigma-Aldrich website product page, the HPA website also contains the same information (www.hpacultures.org.uk). The product number for the cell line is the same on either site.

  5. How do I handle Cell Line frozen cells upon arrival?

    1. Upon receipt, frozen ampules should be transferred directly to vapor phase liquid nitrogen without delay (-135°C) or liquid phase liquid nitrogen if vapor phase is not available. DO NOT use a -80°C freezer as an alternative; this will result in loss of viability.
