Origine de la lignée cellulaire

Human squamous carcinoma

Description de la lignée cellulaire

Derived from an epidermal carcinoma of the vulva taken from an 85 yr old female. The cells carry large numbers of EGF binding sites and is an indicator cell for anti-TGF binding.

Application

A431 Cell Line human has been used:
  • to demonstrate that p38 mitogen-activated protein kinase enhances epidermal growth factor-induced apoptosis in A431 cells[1]
  • to study melanoma-specific CD44 (a tumor marker and transmembrane receptor) alternative splicing pattern in A431 cells[2]
  • to study the anti-cancer activity of antioxidant-rich Cameroonian medicinal plants[3]
Cell cycle studies

Milieu de culture

EMEM (EBSS) + 2mM Glutamine + 1% Non Essential Amino Acids (NEAA) + 10% Foetal Bovine Serum (FBS).

Procédure de repiquage

Split sub-confluent cultures (70-80%) 1:3 to 1:6 i.e. seeding at 2-4x10,000 cells/cm2 using 0.25% trypsin or trypsin/EDTA; 5% CO2; 37°C.

Questions

1–8 sur 8 questions
  1. Can we use A431 cell line for Oral squmaou cell carcinoma research purpose?

    1 réponse
    1. A431 Cell Line human, 85090402, squamous carcinoma, is supplied by ECACC (European Collection of Authenticated Cell Cultures). Please see the Terms and Conditions of Supply at the link below:
      https://www.culturecollections.org.uk/training-and-support/policies/terms-and-conditions-of-supply/

      Utile ?

  2. How many Cell Line cells are in the vial?

    1 réponse
    1. Each vial of cells contains 2-3 x 106 cells in 1 ml of freezing media. This is in a 1.8 mL ampule.  For exceptionally large cells, counts may decrease. Suspension cells, generally smaller cells, may contain as many as 4-5 x 106 cells/vial to assure optimal viability upon thaw.

      Utile ?

  3. How do I handle Cell Line frozen cells upon arrival?

    1 réponse
    1. Upon receipt, frozen ampules should be transferred directly to vapor phase liquid nitrogen without delay (-135°C) or liquid phase liquid nitrogen if vapor phase is not available. DO NOT use a -80°C freezer as an alternative; this will result in loss of viability.

      Utile ?

  4. What passage are my Cell Line cells?

    1 réponse
    1. If the passage of the cells is known, it is listed on the product page at the HPA website (www.hpacultures.org.uk).

      Utile ?

  5. Are my Cell Line cells mycoplasma tested?

    1 réponse
    1. ECACC routinely tests all manufactured cell banks for mycoplasma.

      Utile ?

  6. Why is Vapor Phase Liquid Nitrogen preferred for storage of Cell Line cells?

    1 réponse
    1. If ampules are immersed into liquid phase of liquid nitrogen, it increases the risk of the liquid seeping into the vial. This could lead to problems of cross-contamination and increased risk of the ampule exploding when thawed.

      Utile ?

  7. What medium should I use when I thaw the Cell Line cells?

    1 réponse
    1. The medium for each cell line is listed on the product page. It is specific to each cell line. If not visible on the Sigma-Aldrich website product page, the HPA website also contains the same information (www.hpacultures.org.uk). The product number for the cell line is the same on either site.

      Utile ?

  8. Are the Cell Line cells tested for viral pathogens?

    1 réponse
    1. HPA Cultures does not perform any viral testing on the cell lines.
