General description
Le dithiothréitol (DTT), un agent protecteur des groupements thiol[1], est un excellent réactif pour éviter l'oxydation des groupements SH des protéines. Le DTT en solution est un agent réducteur de pont disulfure puissant et polyvalent.[2] Il réduit les liaisons disulfure en formant un cycle à six chaînons et en initiant un échange thiol-disulfure.
Application
Cette solution de DL-dithiothréitol a été utilisée :
- dans le traitement d'explants intestinaux.
- dans la préparation d'homogénats de cellules de branchies pour des dosages enzymatiques.
- dans l'électrophorèse de protéines pour le western blotting.
- pour l'activité transglutaminase.
- dans un tampon pour échantillon en SDS-PAGE.
- en western blotting, comme constituant d'un tampon de lyse pour le séquençage d'ARN de noyau isolé.
- comme constituant d'un tampon d'extraction de protéines pour la lyse de cellules.
Biochem/physiol Actions
Le DL-dithiothréitol (DTT) est utilisé dans un protocole de rupture des liaisons chimiques qui permet de détacher les bactéries des biofilms. Il est capable de modifier la stabilité structurale du récepteur du GABA(B) (acide gamma-aminobutyrique) en rompant les ponts disulfure situés entre quatre résidus cystéine positionnés dans la structure du récepteur du GABA(B1(a)). Le dithiothréitol (DTT) est largement utilisé dans la synthèse de peptides synthétiques et les structures biochimiques de protéines thiolées. Il peut aussi être utilisé pour étudier divers aspects chimiques des protéines comme leur activité enzymatique ou leur repliement. Le DTT contrôle spécifiquement la réponse de l'échange thiol-disulfure pour réduire complètement les ponts disulfure sub-atomiques intra ou inter-moléculaires dans les biomolécules. Les thiols et le disulfure cyclique du DTT sont les produits de cette réaction.

How can I determine the shelf life / expiration / retest date of this product?

If this product has an expiration or retest date, it will be shown on the Certificate of Analysis (COA, CofA). If there is no retest or expiration date listed on the product's COA, we do not have suitable stability data to determine a shelf life. For these products, the only date on the COA will be the release date; a retest, expiration, or use-by-date will not be displayed.

How is shipping temperature determined? And how is it related to the product storage temperature?

Products may be shipped at a different temperature than the recommended long-term storage temperature. If the product quality is sensitive to short-term exposure to conditions other than the recommended long-term storage, it will be shipped on wet or dry-ice. If the product quality is NOT affected by short-term exposure to conditions other than the recommended long-term storage, it will be shipped at ambient temperature. As shipping routes are configured for minimum transit times, shipping at ambient temperature helps control shipping costs for our customers.
