  1. HI, DOES IT HAVE ANY LACTATE IN THE MEDIUM? IF SO, WHAT CONCENTRATION?

    1. This product does contain Lactate. Please see the full product formulation below. Quantities are in mg/L.

  2. Is the KSOM (MR-121-D) sufficient for embryo culture to the blastocyst stage on its own? In the literature, papers mention using "KSOM supplemented with amino acids." I observed that MR-121-D indicates it has "half amino acids." Essentially, I am trying to understand if individuals are adding extra amino acids to this media, or if the 1/2 amino acids already included are what they are referring to when they say "supplemented with amino acids."

    1. KSOM (MR-121-D) is indeed sufficient for embryo culture up to the blastocyst stage. It is a 1X liquid solution with 1/2 amino acids and Phenol Red. When scientists reference KSOM with 1/2 amino acids, that is the intended composition, and they do not add the remaining 1/2 amino acids.

      For more detailed information and historical context, the article "A brief history of the development of the KSOM family of media" provides insights into the development and usage of KSOM. According to documented literature and the mentioned article, KSOM has been shown to support a high yield of mouse blastocysts capable of developing into fetuses after transfer to surrogate mothers. The medium was notably improved when supplemented with 19 natural amino acids, with the added amino acids present at half the concentration described by Eagle. Dose response studies indicated little difference in the response between full strength and half strength concentrations of the amino acids.
