TMEM173/STING, also known as STING, or Endoplasmic reticulum interferon stimulator, ERIS, Mediator of IRF3 activation, MMITA or Transmembrane protein 173, and encoded by the gene TMEM173/ERIS/MITA/STING, is a facilitator of innate immune signaling that acts as a sensor of cytosolic DNA from bacteria and viruses and promotes the production of type I interferons. STING acts by recognizing and binding cyclic di-GMP (c-di-GMP), a second messenger produced by bacteria, and cyclic GMP-AMP (cGAMP), a messenger produced in response to DNA virus in the cytosol. Upon binding STING is able to activate both NF-kappa-B and IRF3 transcription pathways to induce expression of type I interferon and exert a potent anti-viral state. STING also mediates cell death signaling via the activation of the ERK pathway. STING is found in multiple intracellular locations including the endoplasmic reticulum, plasma cell membrane, mitochondrion outer membrane, and perinuclear regions. STING is expressed in the B-cell lineage both immature and mature B cells but not plasma cells. Mutations in STING cause defects in innate immunity and compromised immune health.
