Specificity

Clone 2C13 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects Protein unc-93 homolog B1 (UNC93B1). It targets an epitope within 19 amino acids from the N-terminal half.

Immunogen

KLH-conjugated linear peptide corresponding to 19 amino acids from the N-terminal half of human Protein unc-93 homolog B1 (UNC93B1).

Application

Quality Control Testing

Evaluated by Western Blotting in HeLa cell lysate.

Western Blotting Analysis: A 1:10,000 dilution of this antibody detected UNC93B1 in HeLa cell lysate.

Tested applications

Western Blotting Analysis: A 1:10,000 dilution from a representative lot detected UNC93B1 in A549 cell lysate and Raji cell membrane extract.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected UNC93B1 in A549 cells.

Immunohistochemistry (Paraffin) Analysis: A 1:1,000 dilution from a representative lot detected UNC93B1 in human small intestine tissue sections.

Affinity Binding Assay: A representative lot of this antibody bound UNC93B1 peptide with a KD of 9.9 x 10-8 in an affinity binding assay.

Flow Cytometry Analysis: 0.1 μg from a representative lot detected UNC93B1 in one million A549 cells.

Target description

Protein unc-93 homolog B1 (UniProt: Q9H1C4; also known as Unc-93B1, hUNC93B1) is encoded by the UNC93B1 (also known as UNC93, UNC93B) gene (Gene ID: 81622) in human. Unc-93B1 is a multi-pass membrane glycoprotein with twelve transmembrane domains. It is a key regulator of nucleic acid (NA)-sensing Toll-like receptors (TLRs) and plays a key role in innate and adaptive immunity. It is highly expressed in antigen-presenting cells. Its expression has been reported in plasmocytoid dendritic cells. Lower levels of expression are observed in heart, kidney, and other tissues. It is localized in endosomes, lysosomes, and endoplasmic reticulum (ER). It re-localizes from ER to endosome and lysosome upon cell-stimulation with CpG dinucleotides. It is thought to guide NA-sensing TLRs (TLR 3, 7, 9) from ER to their respective compartments. UNC-93B1-deficient cells are reported to be unresponsive to TLR3, TLR7, TLR8, and TLR9. Its deficiency is shown to cause a near-complete loss of TLR3 and TLR7 proteins in primary splenic mouse dendritic cells and macrophages. Mutations in UNC93B1 gene are known to cause infection-induced acute encephalopathy and predisposes otherwise healthy subjects to herpes simplex encephalitis due to impaired interferon production. This ZooMAbZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
