Specificity

Clone 2E18 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects Calcitonin gene-related peptide 1. It targets an epitope within 22 amino acids from the active form of CGRP1.

Immunogen

KLH-conjugated linear peptide corresponding to 22 amino acids from human active Calcitonin gene-related peptide 1 (C-terminal region of the uncleaved peptide)

Application

Quality Control Testing

Evaluated by Western Blotting in MCF-7 cell lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected Calcitonin Gene-Related Peptide 1 (CGRP-1) in MCF-7 cell lysate.

Tested Applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected CGRP-1 in PC12 and K562 cells and in Human and mouse liver lysates.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected CGRP-1 in human cerebral cortex and human DRG tissue sections.

Affinity Binding Assay: A representative lot of this antibody bound CGRP-1 peptide with a KD of 1.9 x 10-7 in an affinity binding assay.

Immunocytochemistry Analysis: A 1:1,000 dilution from a representative lot detected CGRP-1 in MCF7 cells.

Anti-CGRP-1, clone 2E18 ZooMAb®, Cat. No. ZRB1267, is a recombinant Rabbit monoclonal antibody that specifically targets CGRP-1 and is tested for use in Affinity Binding Assay, Immunocytochemistry, Immunohistochemistry (Paraffin), and Western Blotting.

Target description

Calcitonin gene-related peptide 1 (UniProt: P06881; also known as Alpha-type CGRP, Calcitonin gene-related peptide I, CGRP-I) is encoded by the CALCA (also known as CALC1) gene (Gene ID: 746) in human. CGRP, a 37 amino acid neuropeptide, is a highly potent and long-acting vasodilator with protective effects in the cardiovascular system. Two forms of CGRP have been described -CGRP-I and CGRP-II (also known as aCGRP and bCGRP) that are synthesized from two distinct genes. CGRP I is the primary neuronal form that is found in the CNS and in the peripheral nervous system and displays higher immunoreactivity than CGRP II. CGRP I is synthesized with a signal peptide (aa 1-25) and two peptides (aa 26-80 and 125-128) that are cleaved off to generate biologically active form (aa 83-119). Following synthesis, it is stored in large, dense-core vesicles within the sensory nerve terminal and is released following neuronal depolarization via calcium-dependent exocytosis. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant mouse monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
